13-residue hydrophobic N-terminal extension (MFPAMPLSSLFVN) joined to the IGF-1 core through a Val-Asn link, plus a Glu3->Arg3 substitution; three intramolecular disulfide bridges as in native IGF-1.
A single-chain polypeptide analogue whose structural binding partner is a receptor tyrosine kinase of the insulin-receptor structural family. The native IGF-1 core retains the three-helix fold and three disulfide bridges of the IGF/insulin family, leaving the receptor-binding region structurally intact. The Glu3->Arg substitution and the hydrophobic 13-residue N-terminal extension alter the charge and steric environment at the N-terminus, lowering affinity for the IGF-binding proteins (IGFBP-1 through -6) that otherwise sequester native IGF-1, while preserving the structural binding determinants of the native core.
