B7-33 is a linear mimetic of the relaxin B-chain that engages a class B1 LGR-family G protein-coupled receptor, contacting its leucine-rich-repeat extracellular domain in a manner analogous to natural relaxin but lacking the disulfide-linked A-chain, producing a compact single-chain format. Structural studies characterize this single-chain binding mode relative to the native two-chain hormone, and its signaling profile in cell-based assays is described as distinct from that of the full relaxin hormone.
