N-terminal tyrosine extension of the hGH 177–191 sequence; intramolecular Cys7–Cys14 disulfide bond forming a cyclic loop.
A disulfide-cyclized 16-residue peptide corresponding to the C-terminal domain of human growth hormone (residues 177–191 plus an N-terminal tyrosine). The N-terminal tyrosine extension and the intramolecular Cys7–Cys14 disulfide bond constrain the backbone into a cyclic loop; NMR conformational analysis of the solution structure identifies type I β-turns in this region. The cyclic loop is distinct from the N-terminal region of the full-length protein, and the tyrosine modification was introduced to improve stability relative to the unmodified C-terminal fragment.
