A linear tridecapeptide analogue of alpha-MSH carrying Nle4 and D-Phe7 substitutions, with N-terminal acetylation and a C-terminal amide. It is characterized as a ligand of a Gs-coupled G-protein-coupled receptor; cryo-EM studies of the receptor–Gs complex have resolved the orthosteric binding pocket and described how a peptide of this class occupies the receptor transmembrane bundle. The Nle4 and D-Phe7 substitutions confer protease resistance, and structural studies have examined how these substitutions affect binding geometry within the orthosteric pocket. The molecule retains the conserved His-Phe-Arg-Trp sequence motif central to its receptor-binding geometry.
