Mechanism & Target Class
A 39-residue synthetic peptide. Positions 2 and 13 are α-aminoisobutyric acid (Aib), conferring resistance to enzymatic degradation by DPP-4; the C20 fatty-diacid chain at Lys20 promotes reversible albumin binding, extending circulatory half-life. Cryo-EM studies have characterized how the N-terminal region engages the orthosteric site of its class B G protein–coupled receptor targets via hydrogen bonds and ionic interactions.
