Mechanism & Target Class
Combines two synthetic peptides with distinct molecular structures. The first component is a 29-residue C-terminally amidated fragment analog corresponding to the N-terminal GRF(1-29) sequence; in structural studies it engages a class B1 (secretin-family) G-protein-coupled receptor, with cryo-EM resolving the peptide contacts across the receptor transmembrane bundle, extracellular loops, and extracellular domain. The second component is a 5-residue pentapeptide (Aib-His-D-2-Nal-D-Phe-Lys-NH2) incorporating non-standard residues — an N-terminal Aib and D-amino-acid substitutions — that confer conformational constraint and protease resistance; in structural studies it occupies the ligand-binding pocket of a class A G-protein-coupled receptor. Both components have been resolved in receptor–ligand complexes at near-atomic resolution, supplying molecular-level reference structures for each peptide's GPCR engagement.
