C-terminal Pro-Gly-Pro tripeptide extension of the ACTH(4-7) core; confers resistance to prolyl endopeptidase degradation; PGP is the principal brain metabolite.
A linear heptapeptide consisting of the ACTH(4-7) core sequence (Met-Glu-His-Phe) extended with a C-terminal Pro-Gly-Pro tripeptide. The Pro-Gly-Pro extension confers resistance to prolyl endopeptidase cleavage relative to the native ACTH(4-10) parent sequence, and the PGP fragment is the predominant brain metabolite identified in distribution studies. In-vitro work has characterized specific, calcium-dependent, reversible binding to a membrane site in rodent brain tissue, consistent with a G-protein-coupled receptor structural context. The methionine residue is oxidation-susceptible, and the histidine and N-terminal residues support copper(II) coordination chemistry studied in neuronal cell models.
